MS15 TFA is a potent, selective AKT PROTAC degrader, effectively inhibiting AKT1, AKT2, and AKT3 with IC50 values of 798 nM, 90 nM, and 544 nM, respectively [1].

体外活性

MS15 (0-10 µM, 24 hours) TFA significantly promotes the degradation of AKT in SW620 cells and MS21 cells with KRAS/BRAF resistance mutations [1]. MS15 (0-10 µM, 5 days) TFA effectively inhibits the proliferation of KRAS mutant SW620 cells [1]. MS15 (1 µM, 1-24 hours) TFA regulates the degradation of AKT through a UPS-dependent mechanism, showing time dependence [1].

体内活性

MS15（75 mg/kg，腹腔注射，单次）经过腹膜给药在小鼠中展示出良好的生物利用度[1]。
